I have an unlocked ATT Galaxy S5 that I have been using for about 2 years on Tmobile. I just got a noticed that ATT had a software update for the phone. If I download and install the update will my phone become locked to ATT again?

I'm pretty sure it does not lock your phone again. Caveat: I don't recall ever getting an upgrade notice as you did.
If it is that old they would allow you to unlock at no charge. There are also free utilities that let you check to see if the phone is locked.
No a phone technician I can tell you it will not lock your phone.
Besides if you phone is out of contract or fully paid for, they unlock it for free.
